Assessing the Potential of Molecular Imaging for Myelin Quantification in Organotypic Cultures

Ex vivo models for the noninvasive study of myelin-related diseases represent an essential tool to understand mechanisms and develop therapies against them. Herein, we assessed potential multimodal imaging traceable myelin-targeting liposomes quantify myelin in organotypic cultures. Methods: MRI testing was used image mouse cerebellar tissue sections Demyelination induced by lysolecithin treatment. Myelin-targeting were synthetized characterized, their capacity tested fluorescence imaging. Results: Imaging freshly excised ranging from 300 µm 1 mm thickness achieved with good contrast between white (WM) gray matter (GM) using T2w MRI. The typical loss stiffness, WM structures, cultures required use diffusion-weighted methods. Designed allowed semiquantitative detection fluorescence, but specificity not consistent assays due unspecific binding liposomes. Conclusions: With respect sensitivity, brain is feasible, nanosystems are a promising solution ex vivo. specificity, fine tuning probe required.
